I think the best thing we can do is try to make the large-file check as robust as possible – and that alone seems to be something of a challenge given all the different and quirky drivers out there. to do otherwise would be much more harmful than helpful. Allowing that check to be readily bypassed would place users' data at grave risk. It would be very dangerous to easily allow the creation of >2 GByte 'single file' disks on filesystems that don't support files larger than 2 GBytes, because the virtual machine would suffer a 'catastrophic virtual disk failure' of sorts when that disk needs to grow beyond 2 GBytes – subsequent writes to new areas of the virtual disk would fail, and the guest OS would most likely crash if its boot volume were affected. The failing ImDisk RAM disk driver has not been written to use the modern Windows Mount Manager, which seems to be why the RAM disks are failing, but that problem should not affect any normal hard disks, pen drives, external HDDs, etc., which would all use the regular Windows driver stack and hence go through the Windows Mount Manager.

We do try to inspect the filesystem type and use a limit appropriate to the known limits of the filesystem, so we should be able to use large files on NTFS (and exFAT, as of Workstation 8, and even up to 4 GBytes on FAT32) as long as the filesystem is handled in the usual way by the Mount Manager component of the Windows host OS.
